Personally I'd dig them out but I guess it depends on how much effort you want/can put in... round up would be a last resort for me especially if you plan on growing veggies etc.
why round up is inactivated with contact with soil, digging up means getting all the root not so easy, theres 2 types of weeds, thistle ,nettles and couch grass(twitch) benefit from systemic spray as it kills the root ,mechanical methods such as hoeing just increase regrowth other weeds can be controlled mechanically very successfully, i grow a lot of veggies so have to deal with lots of weeds

Can yer pull nettles out one by one or is there one of thems long roots that go straight down the middle which makes them grow again?

As a temp solution eye may just pull them so eye can fill the garden bin with as much as eye can as soon as eye can ter make the best use of them coming ter empty it. Then eye can fight them when eye have more time.
Just try not to leave it too long before going back to it or you may find youll end up repeating the job again!

Some people don't like to use chemicals in areas where they want to grow their veg. (May as well go to the shop and buy chemically covered veg). Glyphosate hangs around in the soil for a long time after the weeds have died... it's non specific... loads of reasons not to use it. But everyone to their own.
Besides, for some the reward is more than just growing veg. Digging/ weeding/ tending their plot is just as rewarding as the harvest they get.

glyphosate is non persistent and painting a few leaves isnt going to cause any harm except killing weed and root, i know about growing veggies and different weeds require different solutions, most of my weed control is mechanical but some weeds require more

Based on @Hippo post it sounds like it's going to be a little more than 'painting a few leaves'

Eye have some roundup or resolver - which ever is the blue 3L tub - and had a go at spraying weeds and grass int me block paving. The grass is taking longer ter go but has gone yellow. Weeds have died but they have bigger leaves. Weeds living int rockery int small pebbles have shrunk after sprayin. They had big leaves so it worked faster.

Eye was conscious of using chemicals and trying not to. Eye now have a paving knife L shape hook thing to attack the block paving ter get the bigger weeds out and attack some of the moss int cracks..

While looking at YouTube eye spotted peeps burning weeds ont leaves and anything above soil. Letting me loose with a petrol burner may not end well but eye did see some peeps using an electric hot air gun. That got me thinking. Mine runs at 300 or 600 degrees c.

If eye cut the nettles near the soul and burn the remaining stem... will that kill the root under ground?

Eye have about 5 to 7 square meters if nettles. No rush ter get rid but looking ter make best use of me garden waste wheelie bin collection by making sure it's full every time. Me neighbours were quarter full until eye found out. ;)

Nettles are very persistent!
On my allotment I dug everything out (10ft x 10ft square of 6ft high nettles) then covered the entire area with whatever I could get hold of, cardboard/carpet/ bags etc. Lifting small areas at a time to plant as I did it all over quite a few months... once your plants start to grow it should exclude most weeds though of course you will have to manually weed from there on.

We tried burning with a gas flame thrower a couple of years ago and all it did was make them come back even more determined to infest the place. Yes, it was very hot, even to the point of melting our boot soles when we stood on the ground after the burning was done!!

Eye went ter talk ter me nettles about evicting them un they be as tall as me.
There be more anorl. They int as clever as they think cos the concrete shed base with no shed has stopped them.
