Do I really need to take the gearbox out to do the clutch release bearing?

Nahh you dont.... Folk just take the gearbox out for the fun of it once the clutch is done....and they have time to kill.
All joking aside... some things suck and this is one of them.

Yes.
While you are in there, you may as well clean out the bell housing, fit a new bush and change the clutch.
And a new HD arm.
And check your flywheel is flat ... just done my Defender, it's very satisfying.

New (to me) LT230
I picked up a 'refurbed' transfer box the other day. The suffix is 22D, which is 1:141, the same as a 13D. My question is - what is the difference ?

Good excuse to check the seat box fixings. And handbrake. And propshafts. And rear bearing seal on the engine. Maybe new main bearing shells, all good

I have to admit that’s what I would do, but the point is , the engine and gearbox have to be parted, which effectively what op was asking

If you think of it as removing the body and engine from the gearbox, you feel like you made a massive job much easier.

It could be worse, you could have fitted a new clutch, cleaned and painted the gearbox and fitted a new transfer box to find that the midget rattling the can of spanners is still there and it all needs to come out again .... 
I'm actually a bit worried about that too. Was considering if it's worth putting it in stands so I can run it up before refitting all the floor and seat panels.It could be worse, you could have fitted a new clutch, cleaned and painted the gearbox and fitted a new transfer box to find that the midget rattling the can of spanners is still there and it all needs to come out again ....

Seals have blown on mine, going to tackle it tonight and have it done by Sunday hopefully

No just undo the gearbox, prop, mounts etc then shuffle gearbox back on trolley jack then change clutch and bearing no need to remove seat base.

I have done the job as above when I knew it was just the release bearing that needed changing.
