I have a discovery 300tdi which has a large issue of emptying all the coolant out of the radiator into the expansion tank and then not refilling it, I’ve tried replacing water pumps, it’s has 3 thermostats a new radiator and new coolant hoses and I’ve bled the system several times still no change. Anyone else gone through this and know how to fix

That is how I used to do it, are you refitting header tank cap once water is coming our of the rad bung? then carry on filling via the stat bung, having the header tank cap on when header tank level is correct is the important bit, then carry on filling up via the stat housing bung.
 
No idea how the radiator is emptying into the expansion tank. I've never experienced this. That's not to say it's not possible. I would start with a compression check. Then check the filler cap for blow by. Maybe a sniff test of the coolant. Then drain and remove the from carrier and put a hose pipe through it, flush everything. Including the heater matrix. I had one once blocked up from RTV once. Overused stuff. Also check the auto air bleed valve just by the expansion tank. Check for bubbles in expansion tank when revving. 👍👍
